Resource Notes for Leica DMIRB Inverted Widefield

Setting Up A New user:    (THIS IS A COPY OF WHAT IS AT MAXWELL-212    )

• Logon as user (we need you to do this since your password can not be read).  Alternatively, we your permission, we can reset you password to one known to us and perform the setup.
• Cosmetics:
  • My Computer
    • Display details
    • Tools Folder ---> Options -- View
      • Display Full paths etc.
      • uncheck "Hide Extensions for known file types"
      • uncheck "Remember each folder's view settings"
    • Set desktop background to none
• Remap \\Minsky\your-lab-share to drive letter M:
• Note can not save to this mapped directory or the \\unc due to some weird permission issue.
• Also make shortcuts to user's D:\Users\%username%  and   \\Minsky\your-lab-share
• Create directory:   c:\users\%newuser%
• Edit C:\USER\_Typical\120704-Michaelu3-good.reg and replace all michaelu3 with new user name
• Save as 120704-%newuser%.reg
• Double click on 120704-%newuser%-good.reg
  • Hit OK of course
• Ensure Hamamatsu camera is powered on
• If required ensure MicroPublisher is connected to the USB cable from \\Lister
• Run C-Imaging
• File ---> Current Profile
• Choose Device control tab
• Click on Hamamatsu camera and hit Properties
• Click on Browse
• Select Hamamatsu 1394 ORCA-ERA: s/n: 830743 and press OK
• Press OK twice in subsequent windows
• Optionally make sure paths are good in File ---> Current Profile -- Default File paths tab.  See this example:

• Double click in Camcorder Icon
• In Device Setup tab make sure Capture Resolution is set to 8 bit:
• In camera device setup choose 8 bit
• 
• For low light imaging choose Light Mode: low (for fluorescence)
• For brightfield imaging choose Light Mode: high (for transmitted light)
• In Sensor tab ensure that Fan/Cooling is selected (on)
• In top right of window select Hamamatsu 1394 ORCA-ER
• Start acquiring images with:
  • Live
  • Set Exposure(sec.)
  • Gain
  • Offset (Black level) should be 0 generally
  • Capture 1
  • For color images do not forget to do Color Balance and select a blank area
